目的研究河南益母草野生居群遗传多样性。方法利用SCoT分子标记对位于河南伏牛、太行、大别和桐柏4大山脉的8个益母草居群66个样品进行遗传多样性分析。结果 10对引物共扩增出240条带,其中多态性条带为238条,多态性百分率(PPB)为99.17%。Nei’s遗传多样性指数(H)平均值为0.264 2,Shannon’s多态性信息指数(I)平均值为0.39,遗传分化系数(Gst)为0.294 6,种群间基因流为1.197。Mantel Test分析表明,河南益母草居群间的亲缘关系与它们之间的地理距离有着明显的相关性(r=0.366 3,P<0.05)。结论河南野生益母草种群具有较高的遗传多样性,其遗传距离与地理距离存在明显的相关性。
Objective To study the genetic diversity of native populations of Leonurus in Henan Province. Methods Sixty-six samples from 8 Leonurus populations in four large mountain ranges of Funiu, Taihang, Dabie and Tongbai in Henan were analyzed for their genetic diversity using SCoT markers. Results A total of 240 bands were amplified with 10 pairs of primers, of which 238 were polymorphic bands with the percentage of polymorphism (PPB) of 99.17%. The average value of Nei’s genetic diversity index (H) was 0.264 2, Shannon’s polymorphism information index (I) average was 0.39, genetic differentiation coefficient (Gst) was 0.294 6, and gene flow among populations was 1.197. Mantel Test analysis showed that the genetic relationship among populations of Leonurus in Henan had a significant correlation with the geographical distance between them (r = 0.366 3, P <0.05). Conclusion The population of wild motherwort in Henan Province has high genetic diversity, and its genetic distance and geographical distance are obviously correlated.
