目的探讨不同剂量葛根素注射剂给药后唾液与血液中葛根素浓度的关系。方法雄性Wistar大鼠通过颈静脉给予不同剂量的葛根素注射剂,使用反相高效液相色谱法测定血液及唾液中葛根素的浓度,并采用线性回归分析法评价其相关性。结果给予葛根素注射剂后,血浆中葛根素的浓度随时间呈双指数形式下降,迅速向唾液分布,在颌下腺和腮腺中的唾液浓度相似,差异无统计学意义。不同剂量给药后葛根素在大鼠体内呈线性药动学过程,血液和唾液中葛根素的浓度呈现出显著的相关性。结论可以考虑采用唾液代替血液作为一种生物监测媒介用于葛根素注射剂的治疗药物监测。
Objective To investigate the relationship between saliva and blood puerarin concentration after different doses of puerarin injection. Methods Male Wistar rats were given different doses of puerarin injection through the jugular vein. The concentrations of puerarin in blood and saliva were determined by RP-HPLC. The correlation was evaluated by linear regression analysis. Results After puerarin injection, the concentration of puerarin in plasma decreased with double exponential time and rapidly distributed to saliva. The concentrations of saliva in submandibular gland and parotid gland were similar, but the difference was not statistically significant. After different doses of puerarin in rats with a linear pharmacokinetic process, blood and saliva puerarin concentration showed a significant correlation. CONCLUSIONS Saliva can be used instead of blood as a biological monitoring agent for therapeutic drug monitoring of puerarin injections.
