目的:探讨自身免疫性心肌炎小鼠心肌组织细胞间黏附分子-1(ICAM-1)的表达变化以及三七总皂甙对其的影响。方法:BALB/C雄性小鼠72只,随机均分为对照组(C组)、自身免疫性心肌炎组(A组)和三七总皂甙组(P组)。A组和P组复制自身免疫性心肌炎模型。P组腹腔注射三七总皂甙进行干预。于第14、第21和第30天随机处死各组小鼠8只,留取心脏标本。电镜观察心肌组织超微结构改变,免疫组化法检测心肌组织ICAM-1蛋白表达。结果:A组心肌组织的超微结构损伤严重,P组较A组损伤程度明显减轻;C组心肌组织ICAM-1蛋白表达量极少,A组ICAM-1蛋白表达明显增加,呈动态变化,P组心肌细胞ICAM-1蛋白表达显著低于A组(P<0.05或P<0.01)。结论:自身免疫性心肌炎可诱导心肌组织ICAM-1蛋白的表达,三七总皂甙可能通过下调心肌组织ICAM-1蛋白的表达而减轻心肌损伤。
Objective: To investigate the expression of ICAM-1 in myocardium of autoimmune myocarditis and the effect of Panax notoginseng saponins on it. Methods: Seventy two BALB / C male mice were randomly divided into control group (C group), autoimmune myocarditis group (A group) and Panax notoginseng saponins group (P group). Groups A and P replicate the autoimmune myocarditis model. P group intraperitoneal injection Panax notoginseng intervention. On the 14th, 21st and 30th day, 8 mice in each group were randomly sacrificed and the heart specimens were collected. Ultrastructural changes of myocardial tissue were observed under electron microscope. The expression of ICAM-1 protein in myocardial tissue was detected by immunohistochemistry. Results: The ultrastructural damage of myocardium in group A was serious, and the damage in group P was significantly lessened than that in group A. The expression of ICAM-1 protein in myocardium of group C was extremely low and the expression of ICAM- ICAM-1 protein expression in cardiomyocytes in group P was significantly lower than that in group A (P <0.05 or P <0.01). Conclusion: The expression of ICAM-1 in myocardium can be induced by autoimmune myocarditis. Panax notoginseng saponin may reduce myocardial injury by down-regulating the expression of ICAM-1 in myocardium.
