目的观察慢性丙型肝炎(CHC)患者外周血CD4+T细胞和CD8+T细胞表面程序性死亡配体1(PD-L1)的表达水平以及与HCV RNA载量的关系。方法流式细胞仪检测外周血CD4+T细胞和CD8+T细胞表面PD-L1的表达,荧光定量PCR检测HCV RNA载量,基因芯片检测HCV基因型。结果CHC患者外周血CD4+T细胞和CD8+T细胞表面PD-L1的表达水平较健康人明显上调,与HCV RNA和ALT无相关性。结论PD-L1在CD4+T细胞和CD8+T细胞表面的高表达,抑制免疫细胞清除HCV的能力,导致感染的慢性化。
Objective To observe the expression of PD-L1 on CD4 + T cells and CD8 + T cells in peripheral blood of chronic hepatitis C (CHC) patients and the relationship between the expression of HCV RNA and the load of HCV RNA. Methods Flow cytometry was used to detect the expression of PD-L1 on the surface of CD4 + T cells and CD8 + T cells. HCV RNA was detected by fluorescence quantitative PCR and HCV genotypes were detected by microarray. Results The expression of PD-L1 on CD4 + T cells and CD8 + T cells in peripheral blood of patients with CHC was significantly up-regulated compared with healthy controls, but not related to HCV RNA and ALT. Conclusion The high expression of PD-L1 on the surface of CD4 + T cells and CD8 + T cells inhibits the ability of immune cells to clear HCV and leads to chronic infection.
