目的 :研究星形细胞瘤中端粒酶基因与 P5 3蛋白的表达 ,探讨其在星形细胞瘤诊断和预后中的临床意义。方法 :收集经手术切除及病理证实的星形细胞瘤 86例标本 ,8例正常脑组织作对照。用端粒酶原位杂交检测试剂盒检测组织标本端粒酶基因。用 SABC法检测 P5 3蛋白的表达。结果 :星形细胞瘤中端粒酶基因和 P5 3蛋白的阳性检出率分别为 4 4 .2 % ( 3 8/ 86)和 62 .8% ( 5 4 / 86) ,且随着肿瘤恶性程度的增加 ,端粒酶基因表达强度升高 ( P<0 .0 1) ,端粒酶表达与 P5 3蛋白表达强度无明显相关性 ( P>0 .0 5 )。正常组织中未检测出端粒酶基因。结论 :端粒酶基因与星形细胞瘤的分化程度有关 ,可作为星形细胞瘤恶性程度的重要标记物 ,而 P5 3蛋白可能对端粒酶基因的表达无直接影响
Objective: To study the expression of telomerase gene and P53 protein in astrocytoma and to explore its clinical significance in the diagnosis and prognosis of astrocytoma. Methods: Totally 86 cases of astrocytoma confirmed by surgery and pathology were collected, and 8 normal brain tissues were collected as controls. The telomerase gene was detected by telomerase in situ hybridization kit. The SABC method was used to detect the expression of P53 protein. Results: The positive rates of telomerase gene and P53 protein in astrocytoma were 44.2% (38/86) and 62.8% (54/86), respectively. With the malignant tumor (P <0.01). There was no significant correlation between the expression of telomerase and the expression of P53 protein (P> 0.05). No telomerase gene was detected in normal tissues. CONCLUSION: Telomerase gene is related to the degree of differentiation of astrocytoma and may be used as an important marker of malignancy of astrocytomas, whereas P5 3 protein may not have a direct effect on telomerase gene expression
