目的:建立高效液相色谱法(HPLC)测定新疆软紫草中咖啡酸四聚体含量的方法。方法:采用YMCODSAC18色谱柱(4.6mm×250mm,10μm),流动相:甲醇∶水∶甲酸(29∶71∶0.1),流速:1.0ml/min,检测波长:252nm,柱温:40℃。结果:咖啡酸四聚体进样量在0.5~3.5μg范围内与峰面积线性关系良好,相关系数r=0.9995。平均回收率为101.8%,RSD为2.46%(n=9)。结论:高效液相色谱法适用于新疆软紫草中咖啡酸四聚体含量的测定。
Objective: To establish a HPLC method for the determination of caffeic acid tetramer in Xinjiang Lithospermum. METHODS: A YMCODSAC18 column (4.6 mm×250 mm, 10 μm) was used. The mobile phases were methanol:water:formic acid (29:71:0.1), flow rate: 1.0 ml/min, detection wavelength: 252 nm, and column temperature: 40° C. Results: The calibration curve of caffeic acid tetramer in the range of 0.5~3.5μg showed a good linear relationship with the peak area. The correlation coefficient r=0.9995. The average recovery rate was 101.8% with an RSD of 2.46% (n=9). Conclusion: High performance liquid chromatography is suitable for the determination of caffeic acid tetramer in Xinjiang Lithospermum erythropolis.
