以结扎冠状动脉前降支造成兔心肌梗死后心衰模型,用透射电镜作细胞凋亡形态学观察,免疫组化方法检测bcl 2、Bax,放射免疫法检测ET、NO等,进一步观察中药坎离煎对心衰时心肌细胞凋亡及内皮功能的影响。结果显示,与模型组相比,坎离煎组实验兔心肌细胞bcl 2表达及bcl 2 /Bax比值均显著增加(P<0 05),而开搏通组这两项指标虽有增加,却未显示统计学意义;Bax表达在4组间未显示显著差异。同时,与造模组相比,两药物组心衰兔血浆ET水平显著下降(P<0 05),NO/ET值显著增加(P<0 05),心肌NO显著下降(P<0 05);两药物组间无显著差异(P>0 05)。表明验方坎离煎可能通过增加心肌细胞bcl 2的表达和bcl 2 /Bax比值而减少心衰进程中的心肌细胞凋亡;并可纠正心衰时的内皮功能紊乱,改善血管舒张以减轻心脏后负荷和心衰发展中的心室重构。
The model of heart failure after myocardial infarction in rabbits was obtained by ligation of the anterior descending coronary artery. It was shown that Recipe decoction may reduce cardiomyocyte apoptosis in the process of heart failure by increasing the expression of bcl 2 and bcl 2 /Bax ratio in cardiomyocytes; it can correct endothelial dysfunction in heart failure and improve vasodilation to relieve the heart Ventricular remodeling in the development of stress and heart failure.
