目的探讨人参总皂苷(Total saponin of panax ginseng,TSPG)对神经细胞PC12突起生长以及生长相关蛋白-43(Growth-associated protein-43,GAP-43)表达的影响。方法以0、10、20、40、80 mg/L TSPG作用于体外培养的PC12细胞,相差显微镜观察细胞的形态学改变,透射电镜观察细胞突触连接情况,显微镜测微尺测量突起长度,免疫组织化学和Western blot法检测GAP-43的表达。结果与对照组相比,TSPG处理组分化的PC12细胞直径明显增大,突起长度明显增长,并形成突触连接,GAP-43表达水平增加,以40 mg/L组最为明显。结论 TSPG可诱导PC12细胞发生分化、突起生长延伸,并可上调GAP-43的表达水平。
Objective To investigate the effects of total saponin of panax ginseng (TSPG) on the neurite outgrowth and the expression of growth-associated protein-43 (GAP-43). Methods The cultured PC12 cells were treated with 0, 10, 20, 40, 80 mg / L TSPG. The morphological changes of cells were observed by phase contrast microscopy. The synaptic connections were observed by transmission electron microscopy. Histochemistry and Western blot were used to detect the expression of GAP-43. Results Compared with the control group, the diameter of PC12 cells differentiated by TSPG increased significantly, the length of protuberances increased significantly, and the synaptic connection was formed. The expression of GAP-43 increased, especially in the group of 40 mg / L. Conclusion TSPG can induce the differentiation of PC12 cells and the growth of neurite outgrowth, and up-regulate the expression of GAP-43.
