目的 考察骨定向雌激素———四环素哌嗪雌酚酮 (XW 6 30 )对骨c fos和c jun蛋白表达的影响 ,初步探讨该化合物对骨的作用机制。方法 用小鼠胚胎长骨体外培养模型考察XW6 30对骨生长的作用 ,并采用免疫组织化学方法测定其对软骨静止区、增殖区和肥大区c fos,c jun基因表达产物的影响。结果 XW6 30浓度为 10 -7mol·L-1时 ,长骨骨干和骨总长生长速度显著加快 ,在 10 -9~ 10 -7mol·L-1范围内以剂量依赖方式上调骺板各区c fos和c jun蛋白的表达。 结论 提示XW 6 30可能通过上调软骨细胞c fos和c jun表达而促进软骨内骨形成
Objective To investigate the effect of bone morphogenetic protein-estrogen-tetracycline-piperazine-estrone (XW 6 30) on the expression of c fos and c jun proteins in bone and to explore the mechanism of action of this compound on bone. Methods The effect of XW6 30 on bone growth was investigated by in vitro culture of mouse embryonic long bone. The effects of XW6 30 on the expression of c fos and c jun genes in resting, proliferative and hypertrophic areas were determined by immunohistochemistry. Results When the concentration of XW6 30 was 10 -7 mol·L-1, the growth of long bone and total length of bone significantly accelerated. In the range of 10 -9 -10 -7 mol·L-1, c fos and c jun protein expression. Conclusions XW 6 30 may promote intracartilage bone formation by up-regulating c fos and c jun expression of chondrocytes
