目的:观察和肾络颗粒治疗5/6肾大部切除慢性肾衰竭模型大鼠肾组织增殖细胞核抗原(PCNA)表达的影响,探讨其对肾纤维化的保护作用。方法:采用5/6肾切除模型大鼠,设立正常对照组、模型组、西药福辛普利组、和肾络低剂量组、中剂量组及高剂量组。检测血尿素氮(BUN)、血肌酐(SCr),取残肾组织行HE染色,光镜观察肾组织病理变化,免疫组化检测PCNA的变化。结果:中药组与福辛普利组BUN、SCr明显低于模型组;PCNA在术后表达较正常对照组升高,用药组表达明显低于模型组。结论:和肾络抑制PCNA在肾组织内的过度表达,延缓肾脏纤维化的进程。
Objective: To observe the effect of Shenluo granule on the expression of proliferating cell nuclear antigen (PCNA) in renal tissue of rats with subtotal 5/6 nephrectomy and explore its protective effect on renal fibrosis. Methods: Rats in 5/6 nephrectomy model were established. Normal control group, model group, western medicine fosinopril group, and Shenluo low dose group, middle dose group and high dose group were established. Blood urea nitrogen (BUN) and serum creatinine (SCr) were measured and stained with HE stain. The pathological changes of renal tissue were observed with light microscope. The changes of PCNA were detected by immunohistochemistry. Results: The BUN and SCr in the Chinese medicine group and the fosinopril group were significantly lower than those in the model group. The expression of PCNA was higher than that in the normal control group, and the expression of PCNA in the drug group was significantly lower than that in the model group. CONCLUSION: Shenluo decoction inhibits the over-expression of PCNA in renal tissues and delays the process of renal fibrosis.
