脑尔康及其拆方组对阿尔茨海默病小鼠模型脑内BDNF表达的影响

目的:观察复方中药脑尔康及其拆方组对阿尔茨海默病(Alzheimer’s disease,AD)模型小鼠不同脑区脑源性神经营养因子(brain-derived neurotrophic factor,BDNF)表达的影响。方法:随机将70只昆明小鼠分为7组,每组10只,采用ip氯化铝(AlCl3)溶液100 mg·kg-1ip,隔日1次共40 d,建立AD模型小鼠,于学习记忆成绩测试后,空白组和模型组每天给予生理盐水ig(0.5 mL·d-1);全方组及各拆方组每天分别给予相应药物ig(14.3 g·kg-1·d-1),连续30 d。测定BDNF在各组小鼠右侧皮层、海马、齿状回的阳性表达。结果:①各用药组与模型组比较,全方组及拆方1号组BDNF阳性反应物质表达明显增多(P<0.01);拆方2,3,4号组BDNF表达仅不同程度增多(P<0.05);②各拆方组分别与全方组比较,拆方1号组与全方组BDNF阳性反应物质表达无明显差异;③拆方2,3,4号组与拆方1号组间比较,拆方2号组仅在DG,Cortex区域表达的平均灰度值与拆方1号无明显差异;拆方3,4号组仅在DG区域表达的平均灰度值与拆方1号无明显差异。结论:拆方1号组及脑尔康全方组促进AD模型小鼠脑内BDNF表达作用明显优于其他拆方组,两者疗效相当,但前者药味精简。 OBJECTIVE: To observe the effects of traditional Chinese medicine Naolkang and its disassembled prescriptions on the expression of brain-derived neurotrophic factor (BDNF) in different brain regions of Alzheimer’s disease (AD) model mice. Methods: Seventy Kunming mice were randomly divided into 7 groups (n = 10 in each group). AlCl3 solution (100 mg · kg-1 ip) was used for 40 days every other day to establish AD mice. After the memory test, the blank group and the model group were given normal saline ig (0.5 mL · d-1) daily. The whole group and each split group were given the corresponding drug ig (14.3 g · kg-1 · d-1) For 30 days. The positive expression of BDNF in the right cortex, hippocampus and dentate gyrus in each group was determined. Results: ①Compared with model group, the expression of BDNF-positive reaction in group A and group B was significantly increased (P <0.01); The expression of BDNF in group 2,3,4 was decreased (P <0.05). ②Compared with the whole recipe group, there was no significant difference in the expression of BDNF positive response substance between the first recipe group and the whole recipe group. (3) Between groups 2 and 4, the average gray value only expressed in DG and Cortex area was not significantly different from that of the No. 1 demolition. No significant difference. Conclusion: The effect of BDNF-1 and NaErKang-FangFang on BDNF expression in brain of AD model mice is better than that of other groups.
