目的 :探讨汉防己甲素对肾小球细胞外基质 (ECM )和肾脏转化生长因子 β1 (TGF - β1 )表达改变的影响。方法 :用阿霉素加单侧肾切除制备肾小球硬化动物模型 ,随机分为正常组、肾小球硬化模型组、汉防己甲素组和络活喜组 ,用Northern印迹杂交观察肾TGF - β1 mRNA表达。结果 :两治疗组TGF - β1 mR NA表达均明显下调 ,肾小球ECM也明显减少。结果 :汉防己甲素通过下调TGF - β1 mRNA表达而减少肾小球ECM沉积 ,从而延缓肾小球硬化 ,该药的这一作用与西药络活喜类同。
Objective: To investigate the effects of tetrandrine on the expression of glomerular extracellular matrix (ECM) and transforming growth factor β1 (TGF-β1) in the kidney. METHODS: An animal model of glomerulosclerosis was prepared by unilateral nephrectomy with adriamycin and randomly divided into normal group, glomerulosclerosis model group, tetrandrine group, and Luohuoxi group. The renal TGF was observed by Northern blot hybridization. - β1 mRNA expression. Results: The expression of TGF-β1 mR NA in both treatment groups was significantly down-regulated, and glomerular ECM was also significantly reduced. RESULTS: Tetrandrine inhibited the deposition of glomerular ECM by downregulating the expression of TGF-β1 mRNA, which delayed glomerulosclerosis. This effect was similar to that of western medicine.
