本文报道Ehrlich腹水癌和肺癌带瘤小鼠肝脏中细胞色素P-450(简称P-450)、UDP-葡萄糖醛酸基转移酶(简称UDP-GT)及乳酸脱氢酶(简称LDH)等肿瘤标志酶活性的变化,并在给有机锗化合物(简称Ge)以抑制肿瘤生长时,观察了上述标志酶的变化。Ehrlich腹水癌带瘤小鼠肝微粒体细胞色素P-450含量低于正常对照组,而UDP-GT的“胚胎型”(O-GT)和“新生儿型”(P-GT)活性及肝细胞浆中LDH活性显著增高。在移植肿瘤后,给Ge以抑制肿瘤生长时,小鼠肝脏P-450含量和UDP-GT及LDH活性趋于正常水平。肺癌带瘤小鼠肝脏O-GT和LDH活性均有显著增高,但P-450含量无显著变化。当移植肿瘤后给Ge以抑制肿瘤生长时,肝微粒体O-GT活性趋于正常水平。实验结果表明,Ge对正常小鼠肝脏P-450有抑制作用,而对UDP-GT的胚胎型(O-GT)则有诱导效应。 This article reports tumors such as cytochrome P-450 (abbreviated as P-450), UDP-glucuronosyltransferase (abbreviated as UDP-GT) and lactate dehydrogenase (LDH) in the liver of Ehrlich ascites carcinoma and lung carcinoma bearing mice. Changes in the above-mentioned marker enzymes were observed when changes in enzyme activity were marked and organogermanium compounds (Ge for short) were given to inhibit tumor growth. Liver microsomal cytochrome P-450 content in tumor-bearing mice with Ehrlich ascites carcinoma was lower than that in the normal control group, while UDP-GT’s “embryonic type” (O-GT) and “neonatal type” (P-GT) activity and liver LDH activity in the cytoplasm increased significantly. After transplantation of the tumor, when Ge was administered to inhibit tumor growth, the liver P-450 content and UDP-GT and LDH activity of the mice tended to be normal. The activities of liver O-GT and LDH were significantly increased in lung cancer-bearing mice, but there was no significant change in P-450 content. When Ge was administered to inhibit tumor growth after transplantation of the tumor, liver microsomal O-GT activity tended to be normal. The experimental results show that Ge can inhibit the P-450 in normal mice and induce the embryonic form of UDP-GT (O-GT).
