目的动态观察丹酚酸B(SalB)对脑缺血再灌注小鼠血管内皮细胞分泌功能的影响。方法NIH小鼠随机分为假手术组、模型组、丹酚酸B治疗组和尼莫地平对照组,复制脑缺血再灌注病理模型,并分别在脑缺血30 min再灌注0、30、60 min时测定脑组织一氧化氮(NO)、血浆内皮素(ET)、组织型纤溶酶原激活物(t-PA)、组织型纤溶酶原激活物抑制剂(PAI-1)以及血管性假血友病因子(vWF)的含量。结果模型组不同时点的ET,NO,PAI-1和vWF含量明显升高,而t-PA的含量降低。用丹酚酸B治疗,可调整内皮细胞的分泌功能,尤其是再灌注60 min时可显著降低ET、NO、PAI-1和vWF的含量(P(0.01或P<0.05),同时升高t-PA的含量(P(0.01)。结论丹酚酸B可能通过调整血管内皮细胞分泌舒缩、促凝和抗凝物质的平衡,起到减轻缺血再灌注脑损伤的作用。
Objective To observe the effect of salvianolic acid B (SalB) on the secretion of vascular endothelial cells in cerebral ischemia-reperfusion mice. Methods NIH mice were randomly divided into sham operation group, model group, salvianolic acid B treatment group and nimodipine control group. The pathological models of cerebral ischemia and reperfusion were duplicated, and they were reperfused for 30 min and 30 min after cerebral ischemia. At 60 min, brain tissue was measured for nitric oxide (NO), plasma endothelin (ET), tissue plasminogen activator (t-PA), tissue plasminogen activator inhibitor (PAI-1) and The content of von Willebrand factor (vWF). Results The contents of ET, NO, PAI-1 and vWF in the model group increased significantly at different time points, while the content of t-PA decreased. Treatment with salvianolic acid B can regulate the secretion function of endothelial cells, especially when reperfusion for 60 min can significantly reduce the content of ET, NO, PAI-1, and vWF (P(0.01 or P<0.05), and increase t -PA content (P (0.01). Conclusion Salvianolic acid B may play a role in alleviating cerebral ischemia-reperfusion injury by regulating the balance of vasodilatory, procoagulant and anticoagulant substances in vascular endothelial cells.
