脑络欣通胶囊对脑缺血再灌注模型大鼠血小板聚集及血栓形成的影响

目的:观察脑络欣通胶囊对局灶性脑缺血再灌注损伤大鼠血小板聚集和血栓形成的影响。方法:雄性SD大鼠,随机分为假手术组10只,造模组60只,线栓法制备大脑中动脉闭塞缺血-再灌注(MCAO/R)模型,将造模成功的大鼠随机分为模型组,阿司匹林组(0.010 g·kg-1),脑络欣通高、中、低剂量组(4.500,2.250,1.125 g·kg-1)。透射电子显微镜观察海马神经元超微结构变化,检测各组血清中血栓素B2(TXB2),6-酮前列腺素F1α(6-keto-PGF1α),血小板聚集率及体内血栓形成时间。结果:与假手术组比较,模型组损伤大鼠的神经元的病理形态损坏严重,大鼠血清中TXB2水平明显升高,6-keto-PGF1α水平明显降低,模型组大鼠血小板在1,2,5 min的聚集率和最大聚集率明显升高(P<0.05,P<0.01);与模型组比较,脑络欣通胶囊低、中、高剂量组损伤大鼠的神经元的病理形态学均有明显改善,明显降低大鼠血清中TXB2水平,明显升高6-keto-PGF1α水平,1,2 min时,脑络欣通3个剂量组均明显降低大鼠血小板的聚集率,5 min时,低、高剂量组均明显降低大鼠血小板聚集率,低、中剂量组5 min内的最大聚集率,脑络欣通胶囊低、中剂量组大鼠体内血栓形成时间显著延长(P<0.05,P<0.01)。结论:脑络欣通胶囊具有抑制脑缺血再灌注损伤模型大鼠血小板的聚集和血栓形成,对缺血性脑血管疾病具有一定的保护作用。 Objective: To observe the effect of Naoluotong Capsule on platelet aggregation and thrombosis in rats with focal cerebral ischemia-reperfusion injury. Methods: Male Sprague-Dawley rats were randomly divided into sham operation group (n = 10) and model group (n = 60). MCAO / R model was established by middle cerebral artery occlusion The rats were divided into model group, aspirin group (0.010 g · kg -1) and Naoluoxintong high, medium and low dose groups (4.500,2.250,1.125 g · kg -1). The ultrastructural changes of hippocampal neurons were observed by transmission electron microscopy. The levels of TXB2, 6-keto-PGF1α, platelet aggregation rate and in vivo thrombosis time were detected in each group. Results: Compared with the sham-operation group, the pathological changes of the neurons in the model group were severely damaged. The level of TXB2 in the serum was significantly increased and the level of 6-keto-PGF1α in the model group was significantly decreased (P <0.05, P <0.01). Compared with the model group, the pathomorphology of the neurons in the low, medium and high dose groups of Naoluotong Capsule rats were significantly decreased Significantly decreased TXB2 level in rat serum and significantly increased 6-keto-PGF1α level. After 1,2 min, Naoxintong dose group significantly reduced the platelet aggregation rate in rats, 5 min At the same time, the platelet aggregation rate of rats in both low and high dose groups was significantly reduced, and the maximum aggregation rate within 5 min in low and middle dose groups was significantly increased. The thrombosis time of Naoluotong capsule in low and middle dose groups was significantly prolonged (P < 0.05, P <0.01). CONCLUSION: Naoluotong Capsule can inhibit platelet aggregation and thrombosis in rats with cerebral ischemia-reperfusion injury and have some protective effects on ischemic cerebrovascular diseases.
