目的探讨通过提高冷沉淀比活性来提高FⅧ制剂比活性的方法。方法采用边融化血浆边离心的方式,以及将血浆全部融化并静置后再离心的方式分别提取冷沉淀,然后采用相同的纯化工艺制造FⅧ原液并比较原液的比活性。结果采用边融化边离心的方式获得的冷沉淀经纯化后得到的FⅧ原液比活性最高达到59.4 IU/mg,而采用将血浆全部融化并静置后再离心方式获得冷沉淀经纯化后得到的FⅧ原液比活性最高达208.0 IU/mg。结论采用将血浆全部融化并静置后再离心的方式可以获得比活性较高的冷沉淀,并有利于制造比活性较高的人凝血因子Ⅷ制剂。
Objective To explore the method of increasing the specific activity of FⅧ preparation by increasing the specific activity of cryoprecipitate. METHODS: Centrifugation was performed on the side of thawed plasma, and the cryoprecipitate was extracted by thawing the whole plasma and leaving it to stand for centrifugation, then the same purification process was used to make FⅧ stock solution and compare the specific activity of the stock solution. Results The highest specific activity of FⅧ was obtained by using the method of side melting and centrifugation, and the highest specific activity of FⅧ was 59.4 IU / mg. However, the FⅧ obtained by cryoprecipitation was obtained after all plasma was thawed, The specific activity of the stock solution is up to 208.0 IU / mg.
