目的探讨重组人骨形成蛋白-2成熟肽(rhBMP-2m)对小鼠经6.5~7.5Gyγ射线全身照射引起的放射损伤的治疗作用及其机理。方法小鼠照射后1~6天于腹腔内注射rhBMP-2m生理盐水溶液,每天一次,每次剂量为0.5~2.0mg。观察动物的30天存活情况及几种造血指标。结果治疗组的30天存活率,平均存活时间,CFU-GM集落形成数,骨髓细胞DNA含量,骨髓有核细胞数,脾结节形成数,脾体比及外周血白细胞数均较照射对照组增加,差异有非常显著性(P<0.01)。结论rhBMP-2m对小鼠的放射损伤有治疗作用,其机理可能同诱导造血因子释放,改善造血微环境,促进造血功能恢复有关。
Objective To investigate the therapeutic effect and mechanism of recombinant human bone morphogenetic protein-2 mature peptide (rhBMP-2m) on radiation injury induced by whole body irradiation of mice with γ-rays of 6.5-7.5 Gy. Methods Mice were intraperitoneally injected rhBMP-2m saline solution 1 to 6 days after irradiation, once a day for 0.5 ~ 2.0mg. Observed animals 30 days survival and several hematopoietic indicators. Results The 30-day survival rate, mean survival time, CFU-GM colony formation, DNA content of bone marrow cells, number of bone marrow nucleated cells, number of splenic nodules, splenic body and peripheral blood leukocytes in treatment group were significantly higher than those in control group Increase, the difference is very significant (P <0.01). Conclusion rhBMP-2m has a therapeutic effect on radiation injury in mice. The mechanism may be related to the release of hematopoietic factors, the improvement of hematopoietic microenvironment and the recovery of hematopoietic function.
