目的研究睾酮对人喉癌HEP-2细胞株分裂增殖及端粒酶表达活性的影响。方法采用噻唑蓝(MTT)比色法研究不同浓度睾酮对Hep-2细胞株分裂增殖的影响,用免疫细胞化学检测10-8mol/L睾酮作用48 h细胞雄激素受体及端粒酶的表达活性。结果10-8mol/L睾酮作用48 h,人喉癌细胞Hep-2细胞OD值由作用前的0.150±0.008增至0.281±0.010,其他实验浓度组均不能明显促进Hep-2细胞的分裂增殖。睾酮作用前后Hep-2细胞均强表达雄激素受体,表达率分别为95.73%和93.84%,差别无统计学意义。端粒酶表达率由睾酮作用前的52.35%增至89.21%,端粒酶强阳性细胞爬片由2张增至8张,差异有统计学意义。结论HEP-2细胞株为雄激素敏感型细胞,睾酮可能通过上调端粒酶的表达促进喉癌Hep-2细胞的分裂增殖,这对研究喉癌的激素疗法及开发端粒酶抑制剂有重要的意义。
Objective To study the effects of testosterone on the proliferation and telomerase activity of human laryngeal carcinoma cell line HEP-2. Methods MTT assay was used to study the effect of different concentrations of testosterone on the proliferation and proliferation of Hep-2 cell line. The expression of androgen receptor and telomerase was detected by immunocytochemistry in 48 hours after treated with 10-8mol / L testosterone active.
