目的:研究青礞石对AECOPD痰热证模型大鼠血清CRP、IL-8、TNF-α及肺组织匀浆液中IL-8、TNF-α、ICAM-1、MCP-1含量的影响。方法:采用烟熏加细菌感染结合风热刺激的方法制备AECOPD痰热证模型,分别给予氨茶碱及青礞石混悬液高剂量、混悬液低剂量、水煎液高剂量、水煎液低剂量、药渣高剂量治疗。采用HE染色显微观察各组大鼠肺组织病理学变化,酶联免疫吸附法检测血清及肺组织匀浆液中炎症因子的含量,使用SPSS 18.0统计软件进行数据处理分析。结果:青礞石三种样品的高剂量组大鼠血清中CRP、IL-8、TNF-α及肺组织中IL-8、TNF-α、ICAM-1、MCP-1含量均低于模型组(P<0.01),青礞石混悬液高剂量组的作用略强于药渣高剂量组,但差异无统计学意义。结论:青礞石能有效降低AECOPD痰热证模型大鼠血清及肺组织中炎症因子含量。
OBJECTIVE: To study the effect of corundum on the levels of CRP, IL-8 and TNF-α in serum and IL-8, TNF-α, ICAM-1 and MCP-1 in lung homogenate of AECOPD phlegm-heat syndrome rats. Methods: AECOPD phlegm-heat syndrome model was established by smoking and bacterial infection combined with wind-heat stimulation. High dose of aminophylline and corundum suspensions, low dose of suspension, high dose of decoction, Low-dose liquid, high-dose dregs treatment. The pathological changes of lung tissue of rats in each group were observed by HE staining. The contents of inflammatory cytokines in serum and lung homogenate were detected by enzyme-linked immunosorbent assay (ELISA), and the data were analyzed by SPSS 18.0 software. Results: The contents of CRP, IL-8, TNF-α and IL-8, TNF-α, ICAM-1 and MCP-1 in the serum of the high dose group were lower than those of the model group (P <0.01). The effect of high dose group of corundum suspensions was slightly stronger than that of high dose group, but the difference was not statistically significant. CONCLUSION: Barite can effectively reduce the levels of inflammatory cytokines in serum and lung tissue of AECOPD phlegm-heat syndrome rats.
