目的研究脱毒紫茎泽兰草粉的急性、亚慢性、遗传及生殖毒性并对其做出安全性评价。方法应用急性及亚慢性经口毒性实验、Ames实验、小鼠骨髓嗜多染红细胞微核实验、小鼠睾丸初级精母细胞染色体畸变实验及致畸实验对脱毒紫茎泽兰草粉进行毒理学研究。结果脱毒紫茎泽兰草粉对雌雄性小鼠、大鼠、家兔急性经口半数致死量(LD50)均大于10 000 mg/kg。小鼠骨髓嗜多染红细胞微核实验、小鼠睾丸初级精母细胞染色体畸变实验和Ames实验结果均为阴性。家兔亚慢性毒性实验动物的饲料的摄入量、食物利用率、体质量增长、血常规、尿常规、血生化、脏器系数及组织病理学检查均未见异常。大鼠喂养致畸实验脱毒紫茎泽兰草粉各剂量组孕鼠进食、活动、体质量增长、死胎率、吸收胎率均未见明显影响;胎鼠体质量、身长、尾长与阴性对照组相比,差异无统计学意义(P>0.05);胎鼠外观、骨骼、内脏检查均未发现畸形。结论脱毒紫茎泽兰草粉属实际无毒物质,无遗传及生殖毒性,无致畸作用,长期食用对家兔生长发育无不良影响。
Objective To study the acute, subchronic, genetic and reproductive toxicity of detoxified Eupatorium adenophorum powder and evaluate its safety. Methods Acute and subchronic oral toxicity test, Ames test, mouse bone marrow polychromatic erythrocyte micronucleus test, mouse testis primary spermatocyte chromosome aberration test and teratogenicity test were used to study the toxicity of Eupatorium adenophorum powder the study. Results Acute oral LD50 of E. adenophorum powder was higher than 10 000 mg / kg in male and female mice, rats and rabbits. Mouse bone marrow polychromatic erythrocyte micronucleus test, mouse testicular primary spermatocyte chromosome aberration test and Ames test results were negative. Rabbits subchronic toxicity experimental animal feed intake, food utilization, body weight gain, blood, urine, blood biochemistry, organ coefficient and histopathological examination showed no abnormalities. Rats fed teratogenicity test Pueraria purpurea powder dose groups of pregnant rats eating, activity, body weight gain, stillbirth rate, no significant effect on fetal absorption rate; fetal rat body weight, length, tail length and negative control There was no significant difference between the two groups (P> 0.05). No abnormalities were observed in the appearance, skeletal and visceral examination of fetal rats. Conclusions Detoxification Eupatorium adenophorum is an actual non-toxic substance, non-genetic and reproductive toxicity, no teratogenic effect, long-term consumption of rabbit growth and development without adverse effects.
