目的:探讨16号染色体长臂(16q)金属硫蛋白(metallothionein,MT)基因多态性与中国北方汉族人群2型糖尿病的关系。方法:采用PCR技术和限制性片段长度多态性(RFLP)的方法检测129名糖尿病患者(病例组)和115名正常对照者(对照组)的MT基因家族中的MT2A基因rs10636位点单核苷酸多态性。结果:rs10636 3种基因型GG、GC和CC的频数分布符合Hardy-Weinberg平衡;MT2A的3种基因型GG、GC和CC的频数分布在病例组和对照组中无统计学意义(2χ=0.202,P>0.05),等位基因G和C的频数分布在病例组和对照组中也无统计学意义(2χ=0.136,P>0.05)。结论:MT2A基因rs10636位点可能与2型糖尿病无关。
Objective: To investigate the relationship between metallothionein (MT) gene polymorphism of long arm (16q) on chromosome 16 and type 2 diabetes in Han population of northern China. Methods: PCR was used to detect the rs10636 locus of MT2A gene in MT gene family of 129 diabetic patients (case group) and 115 healthy controls (control group) by restriction fragment length polymorphism (RFLP) Polymorphism. Results: The frequencies of GG, GC and CC in three genotypes of rs10636 were in accordance with Hardy-Weinberg equilibrium. The frequency distribution of GG, GC and CC in the three genotypes of MT2A was not statistically significant (2χ = 0.202 , P> 0.05). The frequency distribution of alleles G and C was also not statistically significant in cases and controls (2χ = 0.136, P> 0.05). Conclusion: The rs10636 locus of MT2A gene may not be associated with type 2 diabetes mellitus.
