目的 在新生牛坐骨神经提取对背根神经节 (dorsalrootganglia,DRG)细胞存活和突起生长有抑制活性的物质 .方法 原代培养 DRG细胞 ,实验组分别以不同浓度加入前一实验证实对 PC12细胞有抑制活性 ,Mr5 0 0 0~ 10 0 0 0之间的新生牛坐骨神经提取液 ,对照组中加入培养液 . 48h后进行MTT细胞活性测定、总蛋白含量测定以及观察细胞突起生长情况 .结果 实验组加入提取液蛋白浓度为 2 0 m g· L- 1 ,40mg· L- 1时 ,DRG细胞的活性、总蛋白含量以及突起的生长无明显变化 ;当提取液的浓度为 80 mg· L- 1 ,16 0 mg· L- 1时 ,DRG细胞胞体皱缩 ,突起生长缓慢、停滞 ,细胞活性和总蛋白含量明显减低 .对照组的 DRG细胞生长良好 .结论 新生牛坐骨神经内存在一种对体外培养的 DRG细胞存活及突起生长有抑制作用的物质 .
OBJECTIVE: To extract substances that inhibit the survival and growth of dorsal root ganglia (DRG) cells in neonatal sciatic nerve.Methods DRG cells were cultured in primary culture, Activity, the newborn sciatic nerve extract of Mr 500 ~ 100 000, the control group was added to the culture medium 48h after MTT cell activity determination, determination of total protein content and the observation of cell growth.Results The experimental group was added When the concentration of extract protein was 20 mg · L-1, 40 mg · L-1, the activity of DRG cells, the total protein content and the growth of protuberant cells did not change obviously. When the concentration of extract was 80 mg · L-1, 16 At 0 mg · L -1, the cell bodies of DRG cells shrank, the neurites grew slowly, stunted, the cell activity and the total protein content decreased obviously.Conclusion DRG cells grow well in the control group.Conclusion There is a kind of in vitro DRG Cell survival and growth inhibition of the growth of substances.
