[目的]探索木瓜蛋白酶提取茶树菇多糖的最佳反应条件。[方法]利用酶能够破坏细胞的组织结构的原理,选用木瓜蛋白酶酶解细胞获取多糖。同时考虑到影响酶作用的几个因素,利用正交法研究了木瓜蛋白酶在4个因素(酶解温度、酶解时间、酶浓度和料液比)3水平下的提取效果。[结果]正交试验确定了木瓜蛋白酶提取茶树菇多糖的得率最高的是:酶解温度为45℃、酶解时间2 h、酶解浓度为2%、料液比1∶80,在该提取条件下,茶树菇多糖的得率为5.5%。[结论]正交试验结果确定了木瓜蛋白酶法提取茶树菇多糖的最佳反应条件。
[Objective] The research aimed to explore the optimal reaction conditions for extracting papyrus polysaccharide from papain by papain. [Method] Using enzyme can destroy the principle of cell structure, choose papain to obtain polysaccharides by enzymatic hydrolysis. At the same time, taking into account several factors that affect the enzyme, the orthogonal test was used to study the extraction efficiency of papain at 4 levels (enzymolysis temperature, enzymolysis time, enzyme concentration and solid-liquid ratio). [Result] The results of orthogonal experiment showed that the highest yield of polysaccharides extracted by papain was: enzymolysis temperature 45 ℃, enzymolysis time 2 h, enzymatic hydrolysis concentration 2%, solid - liquid ratio 1:80, Extraction conditions, Chaxingu polysaccharide yield of 5.5%. [Conclusion] The optimal reaction conditions for the extraction of polysaccharides from Camellia sinensis by papain were determined by orthogonal test.
