为了探讨葛根异黄酮对野百合碱诱导的肺动脉高压大鼠血管内皮生长因子(vascular endothelial growth factor,VEGF)表达的影响,我们将45只雄性SD大鼠随机分成正常组对照、野百合碱组、葛根异黄酮治疗组。监测血流动力学变化及观察肺小动脉结构重塑,Western Blot检测肺组织VEGF的分布及表达。结果显示,野百合碱组大鼠平均肺动脉压(mean pulmonary arterial pressure,MPAP)较正常组明显升高(p<0.05),葛根异黄酮治疗组较野百合碱组MPAP明显降低(p<0.05),肺小动脉重塑显著改善。野百合碱组大鼠肺组织VEGF的表达较正常组及葛根异黄酮治疗组明显下调(p<0.05),葛根异黄酮治疗组较野百合碱组VEGF的表达明显上调(p<0.05)。本研究表明葛根异黄酮能减缓肺动脉高压的发生发展,其机制可能与其上调VEGF表达有关。
To investigate the effect of isoflavone on the expression of vascular endothelial growth factor (VEGF) in monocrotaline-induced pulmonary hypertension rats, 45 male Sprague-Dawley rats were randomly divided into normal control group, monocrotaline group, Pueraria isoflavones treatment group. The changes of hemodynamics and remodeling of pulmonary arterioles were observed. The distribution and expression of VEGF in lung tissue were detected by Western Blot. The results showed that the average pulmonary arterial pressure (MPAP) in the monocrotaline group was significantly higher than that in the normal group (p <0.05), and the MPAP in the Pueraria lobata group was significantly lower than that in the monocrotaline group (p <0.05) Pulmonary arteriolar remodeling was significantly improved. The expression of VEGF in lungs of monocrotaline group was significantly lower than that of normal group and isoflavone group (p <0.05), and the expression of VEGF in monocrotaline group was significantly higher than that of monocrotaline group (p <0.05). This study shows that Pueraria isoflavones can slow the development of pulmonary hypertension, the mechanism may be related to its up-regulation of VEGF expression.
