目的 研究结核分支杆菌 MPT64、ESAT6和Ag85A DNA疫苗的抗结核作用。方法 用结核分支杆菌H37 Rv腹腔内注射BALB/c。小鼠2个月后,将小鼠随机分为8组,分别用生理盐水(A)、载体质粒(B)、卡介苗(C)、微卡菌苗(D)、MPT64 DNA疫苗(E)、ESAT6 DNA疫苗(F)、Ag85A DNA疫苗(G)和联合DNA疫苗(H)免疫小鼠,ELISA法检测血清抗体。免疫治疗2个月和5个月时,分别取肺、肝和脾脏观察病理改变并称取重量,作菌落计数、肺组织涂片及脾淋巴细胞转化试验。结果 各DNA疫苗组抗原特异性抗体均不同程度升高。免疫治疗2月时,各组鼠脾淋巴细胞转化率差异无显著意义,各治疗组肺组织涂片细菌数和肺菌落数均比对照组不同程度地减少,只有D组的肺菌落数和E组脾菌落数显著低于对照组,C、E和B组的肺病变指数较低。免疫治疗5个月时,各治疗组鼠脾淋巴细胞转化率均显著增高,各组肺、脾细菌数无显著差别,G和D组的肺未见明显病变。结论DNA疫苗具有一定的抗结核治疗作用。
Objective To study the anti-TB effects of Mycobacterium tuberculosis MPT64, ESAT6 and Ag85A DNA vaccines. Methods BALB / c was injected intraperitoneally with Mycobacterium tuberculosis H37 Rv. After 2 months, mice were randomly divided into 8 groups and treated with saline (A), vector plasmid (B), BCG (C), Mika vaccine (D), MPT64 DNA vaccine ESAT6 DNA vaccine (F), Ag85A DNA vaccine (G), and combined DNA vaccine (H). Serum antibodies were detected by ELISA. Immunotherapy 2 months and 5 months, respectively, to take the lung, liver and spleen were observed pathological changes and weighed, for colony count, lung tissue smear and spleen lymphocyte transformation test. Results DNA vaccine group antigen-specific antibodies were increased to varying degrees. At 2 months of immunotherapy, there was no significant difference in the rate of spleen lymphocyte transformation between the two groups. The number of bacteria and the number of lung colonies in lung tissue in each treatment group decreased to some extent than the control group. Only the numbers of lung colonies and E The number of spleen colonies in the group was significantly lower than that in the control group, and the lung index in group C, E and B was lower. At 5 months after immunotherapy, the conversion rate of spleen lymphocytes in each treatment group was significantly increased, there was no significant difference in the number of bacteria in lung and spleen of each treatment group, and no obvious pathological changes in the lungs of G and D groups. Conclusion DNA vaccine has some anti-TB therapeutic effect.
