[目的]采用蛋白质组学技术鉴定肝癌细胞株HepG2的G1期细胞内的不均一性核糖核蛋白A2/B1(hnRNPA2/B1)蛋白。[方法]提取肝癌细胞株HepG2的G1期细胞的蛋白质,采用双向电泳方法分离蛋白质,应用图像扫描仪及质谱(MALDI-TOF-MS)分析鉴定HepG2的G1期细胞表达的蛋白质。[结果]采用双向电泳技术分离,并用质谱成功鉴定出HepG2的G1期细胞的hnRNPA2/B1。[结论]肝癌细胞株HepG2的G1期细胞内的hnRN-PA2/B1蛋白的成功鉴定,为探讨其在肝癌细胞生长和增殖中的作用奠定了基础,并为临床上肿瘤的治疗提供参考依据。
[Objective] To identify the heterogeneous ribonucleoprotein A2 / B1 (hnRNPA2 / B1) protein in G1 phase cells of HepG2 cell line by proteomics. [Method] The protein of G1 phase of HepG2 cell line was extracted. The protein was separated by two - dimensional gel electrophoresis. The protein of G1 phase of HepG2 was identified by MALDI-TOF-MS. [Result] The hnRNPA2 / B1 of G1 phase cells of HepG2 were successfully identified by two-dimensional electrophoresis and mass spectrometry. [Conclusion] The successful identification of hnRN-PA2 / B1 protein in G1 phase cells of HepG2 cell line lays the foundation for exploring the role of hnRN-PA2 / B1 protein in the growth and proliferation of hepatocellular carcinoma cells and provides a reference for the clinical treatment of tumors.
