目的:观察A-RDMJ对小鼠涎腺放射性损伤后唾液生化变化的影响。为A-RDMJ的进一步改进提供实验基础。方法:将120只昆明小鼠随机分成对照组、单纯放射组、放射前给药组、放射后给药组、放射前后均给药组。除对照组外,其它各组以直线加速器7.5Gy一次性照射小鼠头颈部。给药各组在各时间点腹腔注射A-RDMJ1g/kg,对照组腹腔注射等量生理盐水。放射前后测量各组小鼠唾液钠钾离子浓度、唾液pH、唾液淀粉酶浓度,比较各组差异,并对同一只小鼠照射前后唾液标本进行SDS-PAGE电泳,比较照射前后蛋白组成变化。结果:各实验组照射后唾液钠离子浓度升高,钾离子浓度没有变化。单纯照射组和放射后给药组,两组比较无显著性差异,p﹥0.05,钠离子浓度高于单纯放射组、放射前后给药组,两组比较无显著性差异,p﹥0.05。照射后实验各组唾液中淀粉酶浓度、pH明显下降,放射前给药组和放射前后给药组唾液淀粉酶浓度、下降百分率低于单纯照射组、放射后给药组,两组没有显著性差pH异,﹥0.05。单纯放射组小鼠在接受射线照射后23.5KD条带蛋白和对照组相比大量下降,而给药各组的该蛋白则都p较单纯放射组含量高,不同程度地接近对照组的蛋白浓度。结论:A-RDMJ可减少小鼠涎腺放射性损伤造成的唾液生化指标变化,保护涎腺。
Objective: To observe the effect of A-RDMJ on the salivary biochemical changes after salivary gland irradiation in mice. Provide experimental basis for the further improvement of A-RDMJ. Methods: 120 Kunming mice were randomly divided into control group, radiation group, radiation group, radiation group and radiation group. In addition to the control group, the other groups with a linear accelerator 7.5Gy one-time irradiation in mice head and neck. Administration of each group at each time point by intraperitoneal injection of A-RDMJ1g / kg, control group intraperitoneal injection of the same amount of saline. Before and after radiotherapy, the concentrations of sodium and potassium in saliva, saliva pH and salivary amylase in each group were measured, and the differences among groups were compared. SDS-PAGE electrophoresis before and after the same mouse irradiation was used to compare the changes of protein composition before and after irradiation. Results: The concentration of sodium in saliva increased after irradiation in each experimental group, while the concentration of potassium did not change. There was no significant difference between the two groups (p> 0.05). The sodium ion concentration was higher than that of the radiotherapy alone group and the radiotherapy group. There was no significant difference between the two groups (p> 0.05). After the irradiation, the salivary amylase concentration and pH of the experimental groups decreased significantly. The concentrations of salivary amylase in the pre-irradiation group and the pre-irradiation group were lower than those in the pure irradiation group and the irradiated group. There was no significant difference between the two groups pH difference,> 0.05. Compared with the control group, the 23.5KD banding protein in the radiotherapy group significantly decreased compared with that in the control group, while the protein in each group was higher than that in the radiotherapy group alone, and was close to the protein concentration in the control group . Conclusion: A-RDMJ can reduce salivary biochemical changes caused by salivary gland radiation and protect the salivary glands.
