采用生物素—抗生物素蛋白DCS体系(biotin-avidin DCS system)间接免疫荧光新技术,以176小鼠抗人A431细胞的表皮生长因子受体McAb进行表皮生长因子受体在小鼠睾丸内免疫组织化学定位的研究。在体实验结果显示,小鼠睾丸曲细精管的精原细胞和支持细胞内可见斑点状荧光,其它各级生精细胞均未见荧光。离体实验结果显示,体外培养的小鼠支持细胞内有相当强的荧光。证明精原细胞和支持细胞是表皮生长因子的靶细胞。
Indirect immunofluorescent staining with biotin-avidin DCS system and epidermal growth factor receptor McAb of 176 mouse anti-human A431 cells were used to immunize mice in the testicular Study of histochemical localization. In vivo experiments showed that the mouse testicular seminiferous tubule spermatogonia and supporting cells can be seen in the spot-like fluorescence, and other levels of germ cells at all levels did not see the fluorescence. In vitro experiments showed that in vitro cultured mouse cells have quite strong fluorescence. It is demonstrated that spermatogonia and supporting cells are epidermal growth factor target cells.
