目的:研究伊马替尼对大鼠C6胶质瘤细胞增殖的影响。方法:用不同浓度的伊马替尼(0.039,0.078,0.156,5,10,15,17.5,20μmol.L-1)分别处理C6胶质瘤细胞,作用时间分别为24,48,72,96h,采用四甲基偶氮唑盐(MTT)比色法检测伊马替尼用药后C6胶质瘤细胞的增殖情况。结果:伊马替尼对C6胶质瘤细胞的作用呈时间-剂量依赖性,孵育96h、20μmo.L-1的伊马替尼对C6细胞的抑制作用最强,抑制率接近90%;24,48,72,96h的IC50分别为26.10,21.46,13.32,11.84μmol.L-1。结论:伊马替尼对C6胶质瘤细胞的增殖具有显著的抑制作用。
Objective: To study the effect of imatinib on the proliferation of rat C6 glioma cells. Methods: C6 glioma cells were treated with different concentrations of imatinib (0.039,0.078,0.156,5,10,15,17.5,20μmol.L-1) for 24,48,72,96h , The proliferation of C6 glioma cells after imatinib treatment was detected by MTT assay. RESULTS: Imatinib had a time-and dose-dependent effect on C6 glioma cells. After 96 hours incubation, imatinib at 20μmo.L-1 had the strongest inhibitory effect on C6 cells with the inhibition rate reaching 90%. 24 The IC50 of 48, 72, 96h were 26.10,21.46,13.32,11.84μmol.L-1 respectively. Conclusion: Imatinib has a significant inhibitory effect on the proliferation of C6 glioma cells.
