目的探讨微小RNA-205(miRNA-205)在非小细胞肺癌(NSCLC)组织和血浆中的表达和临床意义。方法用实时定量PCR法测定31例NSCLC患者(试验组)及10例肺良性病变患者(对照组)支气管镜活检组织和血浆中miRNA-205的表达量,分析其与各临床病理资料之间关系,评价miRNA-205在NSCLC的诊断价值。结果试验组活检组织及血浆miRNA-205表达量均高于对照组(P<0.05)。miRNA-205在鳞癌组织中的表达高于腺癌(P<0.05)。miRNA-205的表达量与NSCLC患者临床病理特征无明显相关(rs=0.255,P=0.113)。ROC曲线分析显示,取界值0.932时,组织miRNA-205表达诊断NSCLC的灵敏度和特异度分别为81.3%和83.3%;取临界值0.610时,血浆miRNA-205表达诊断NSCLC的灵敏度和特异度分别为81.3%和84.2%。结论 miRNA-205在NSCLC组织和血浆均有较高的表达,可能会成为诊断NSCLC,尤其是鳞癌的参考指标。
Objective To investigate the expression of microRNA-205 (miRNA-205) in non-small cell lung cancer (NSCLC) tissues and plasma and its clinical significance. Methods The expression of miRNA-205 in bronchial biopsies and plasma from 31 patients with NSCLC (experimental group) and 10 patients with benign lung disease (control group) was detected by real-time PCR. The relationship between miRNA-205 and clinicopathological data , To evaluate the diagnostic value of miRNA-205 in NSCLC. Results The expression levels of miRNA-205 in biopsy tissues and plasma in experimental group were higher than those in control group (P <0.05). The expression of miRNA-205 in squamous cell carcinoma was higher than that in adenocarcinoma (P <0.05). There was no significant correlation between the expression of miRNA-205 and the clinicopathological features of NSCLC patients (rs = 0.255, P = 0.113). ROC curve analysis showed that the sensitivity and specificity of tissue miRNA-205 expression in diagnosing NSCLC were 81.3% and 83.3% respectively at the cutoff of 0.932. When the cutoff value was 0.610, the sensitivity and specificity of plasma miRNA-205 expression in diagnosing NSCLC were 81.3% and 84.2%. Conclusion The high expression of miRNA-205 in both NSCLC tissues and plasma may be a reference for diagnosis of NSCLC, especially squamous cell carcinoma.
