目的 了解角蛋白 14 (K14 )在人胎儿表皮发育中的变化及意义。方法 早、中、晚期胎儿皮肤做冰冻切片 ,进行K14的免疫细胞化学染色 (Immunocytochemistry ,ICC)和原位杂交染色 (insituhybridization ,ISH) ,通过图像分析和统计学处理分析K14的变化及规律。结果 人胚胎 12周表皮已表达K14 ,中期表达量最高 ,晚期也有较高表达 ,但主要局限于表皮基底层细胞。结论 胎儿表皮基底层角朊细胞 (KC)表达K14 ,对维持KC形态 ,防止细胞溶解起重要作用
Objective To understand the change and significance of keratin 14 (K14) in the development of human fetal epidermis. Methods Fetal skin of early, middle and late stage was frozen and sectioned. Immunocytochemistry (KC), immunocytochemistry (ICC) and in situ hybridization staining (ISH) were performed. The changes and regularities of K14 were analyzed by image analysis and statistical analysis. Results The human embryonic 12 weeks epidermis had expressed K14, the mid-term expression was the highest, but also higher in the late stage, but it was mainly confined to the epidermal basal cells. Conclusion The expression of K14 in fetal epidermal keratinocytes (KCs) may play an important role in maintaining KC morphology and preventing cell lysis
