目的 探讨Fas、FasL、bcl 2在大肠癌中表达及意义。方法 用免疫组化S P法检测 15例大肠腺瘤、4 0例大肠腺癌及 15例大肠正常组织Fas、FasL、bcl 2蛋白表达 ,以TUNEL法测定细胞凋亡。结果 腺瘤和腺癌凋亡指数高于正常组织 ,腺瘤组高于腺癌组 (P <0 .0 5 )。在大肠正常组织、腺瘤、腺癌中Fas阳性率下降 ,FasL阳性率上升 ,有显著性差异 (P <0 .0 5 )。腺瘤组和腺癌组bcl 2表达高于正常组 (P <0 .0 5 )。bcl 2阳性组FasL阳性率高于bcl 2阴性组 (P <0 .0 5 )。结论 瘤细胞Fas FasL表达异常使肿瘤逃脱机体自身免疫攻击 ,促使肿瘤的发生、发展 ,对预测大肠癌的预后有重要参考价值。bcl 2高表达是癌变过程中的早期事件 ,有利肿瘤的早期诊断。
Objective To investigate the expression and significance of Fas, FasL and bcl2 in colorectal cancer. Methods The expressions of Fas, FasL and bcl 2 protein in 15 cases of colorectal adenoma, 40 cases of colorectal adenocarcinoma and 15 cases of large intestine were detected by immunohistochemical SP method. The apoptosis was detected by TUNEL method. Results Apoptosis index of adenoma and adenocarcinoma was higher than that of normal tissue, adenoma group was higher than adenocarcinoma group (P <0.05). In normal colorectal tissues, adenomas and adenocarcinomas, the positive rate of Fas decreased and the positive rate of FasL increased, with significant difference (P <0.05). The expression of bcl-2 in adenoma group and adenocarcinoma group was higher than that in normal group (P <0.05). The positive rate of FasL in bcl 2 positive group was higher than that in bcl 2 negative group (P <0.05). Conclusion The abnormal expression of Fas FasL in tumor cells makes the tumor escape the autoimmune attack of the body to promote the occurrence and development of the tumor and has an important reference value for predicting the prognosis of colorectal cancer. High expression of bcl 2 is an early event in the process of carcinogenesis, which favors the early diagnosis of tumors.
