目的:探究解毒胶囊对乳腺增生大鼠乳腺组织的形态、fasl和fas蛋白表达、caapase3mRNA表达的影响。方法:选取40只12周龄SD雌性未孕大鼠随机分为4组(正常组、模型组、方药组、三苯氧胺组),每组10只,造模成功后,经药物治疗4周,用HE染色观察各组乳腺组织形态,用免疫组化技术检测各组大鼠乳腺组织fas和fasl蛋白表达情况,用QPCR技术检测各组大鼠caspase3mRNA表达情况。结果:HE染色结果显示:模型组与正常组相比,乳腺腺泡腔面积变大;方药组和三本氧胺组与模型组相比,乳腺腺泡腔面积有明显减小。免疫组化结果显示正常组fas呈强阳性表达,模型组呈阴性或弱阳性表达,方药组和三苯氧胺组呈强阳性表达。而fasl表达情况与fas表达情况相反,正常组fasl弱阳性表达,模型组呈强阳性表达,方药组和三苯氧氨胺组呈弱阳性表达。QPCR结果显示模型组caspase3与正常组相比呈低表达(P<0.1),治疗组、三本氧胺组与模型组相比呈高表达(P<0.01),方药组和三苯氧胺组比起来没有统计学意义(P>0.05)。结论:解毒胶囊能够干预乳腺增生大鼠的乳腺组织形态、乳腺组织的fasl fas蛋白表达、乳腺组织caspase3mRNA表达,对乳腺增生病有治疗作用。
Objective: To investigate the effect of Jiedu capsule on breast tissue morphology, fasl and fas protein expression and caapase3 mRNA expression in mammary gland hyperplasia rats. Methods: Forty non-pregnant SD female rats of 12 weeks old were randomly divided into 4 groups (normal group, model group, prescription group and tamoxifen group) with 10 rats in each group. After successful treatment, The morphological changes of mammary gland were observed by HE staining. The expression of fas and fasl protein in mammary gland was detected by immunohistochemistry. The expression of caspase 3 mRNA in each group was detected by QPCR. Results: The results of HE staining showed that the area of mammary gland acinar became larger than that of the normal group. Compared with the model group, the area of mammary gland acinar area was significantly reduced in the prescription group and the triamcinolone group. Immunohistochemistry results showed that the fas group showed a strong positive expression in the normal group and a negative or weakly positive expression in the model group, and strong positive expression in the prescription group and the tamoxifen group. However, the expression of fasl was opposite to the expression of fas. The expression of fasl in normal group was weakly positive and the expression of fasl was strongly positive in the model group. The positive expression was weak in the prescription group and the tamoxifen group. QPCR results showed that the expression of caspase 3 in model group was significantly lower than that in normal group (P <0.1), and the expression of caspase 3 in model group was significantly higher than that of model group (P <0.01) Statistical significance (P> 0.05). Conclusion: Jiedu capsule can interfere the mammary gland morphology, fasl fas protein expression in breast tissue and caspase3 mRNA expression in mammary gland hyperplasia rats, which may have a therapeutic effect on mammary gland hyperplasia.
