利用已知植物的蛋白激酶及抗病基因氨基酸序列,设计含激酶保守功能域的简并引物,运用mRNA差异显示技术显示经病原菌诱导的抗性品系(IR26)及感病品系(金刚30)愈伤组织mRNA表达差异,并对差异片段分子克隆.Northern杂交结果提示克隆片段确受白叶枯病病菌诱导表达且该基因仅在抗性品系IR26中存在高水平表达.
Degenerate primers containing conserved domains of kinases were designed using amino acid sequences of protein kinases and disease-resistance genes of known plants. The differential display of mRNA was used to show that the pathogen-induced resistance strain (IR26) and susceptible strain (Vajray 30) Wound tissue mRNA expression differences, and molecular cloning of differential fragments. Northern blot results suggested that the cloned fragment was indeed induced by the bacterial blight and the gene was highly expressed only in the resistant strain IR26.
