目的:探讨灵芪胶囊对H_(22)荷瘤小鼠的抑瘤作用及其抑瘤作用机制。方法:采用体内实验法,于无菌条件下抽取传代7d、生长良好的H_(22)荷瘤小鼠腹水,无菌0.9%氯化钠溶液1:3稀释(约含瘤细胞2×10~6/ml),按0.2ml/只接种于消毒后的小鼠右前肢腋部皮下,制成肿瘤模型,灌胃给药灵芪胶囊11d后,称取小鼠体重,脱椎处死小鼠,取脾脏及胸腺称重并计算胸腺指数和脾脏指数。用免疫组化法检测各组凋亡相关基因突变型p53和bcl-2基因的表达。结果:灵芪胶囊在明显抑制肿瘤生长的同时不降低体重,在一定剂量对荷瘤动物的免疫器官指数无明显影响;灵芪胶囊三个剂量组突变型p53基因和bcl-2基因的表达均低于模型组。结论:灵芪胶囊对H_(22)荷瘤小鼠有明显抑瘤作用,其机制可能与抑制突变型p53基因和bcl-2基因表达有关。
Objective: To investigate the anti-tumor effect of Lingqi Capsule on H22-bearing mice and its anti-tumor mechanism. METHODS: In vivo tests were used to extract ascites from well-cultivated H_22 tumor-bearing mice that were passaged for 7 days under aseptic conditions. A 1:3 dilution of sterile 0.9% sodium chloride solution was used (approximately 2 × 10 cells with tumor cells). 6/ml) 0.2 ml/inoculated subcutaneously in the right anterior iliac crest of the sterilized mouse to make a tumor model. After 11 days of oral administration of Lingqi capsules, mice were weighed and sacrificed. Spleen and thymus were weighed and thymus index and spleen index were calculated. Immunohistochemistry was used to detect the expression of apoptosis-related gene mutant p53 and bcl-2 genes in each group. RESULTS: Lingqi Capsules did not reduce body weight while significantly inhibiting tumor growth, and had no significant effect on immune organ index of tumor-bearing animals at certain doses; Expression of mutant p53 and bcl-2 genes in Lingzhi capsules at three dose groups Lower than the model group. Conclusion: Lingqi capsules have significant anti-tumor effect on H22 tumor-bearing mice. The mechanism may be related to the inhibition of mutant p53 gene and bcl-2 gene expression.
