目的:建立人结肠腺癌细胞系(Caco-2)细胞模型并对其药物研究的方法进行初步探讨。方法:通过测定跨细胞膜电阻(TEER)、荧光素通透量及碱性磷酸酶(AKP)评价Caco-2细胞模型,用MTT法考察HBSS缓冲液在不同pH值、不同温度及不同时间条件下对Caco-2细胞的毒性。结果:各项指标的测定值符合要求,表明本实验建立的Caco-2细胞模型的完整性、紧密性和通透性良好;进行药物Caco-2细胞吸收潜性研究时,药物作用4 h之内对Caco-2细胞模型没有影响。结论:建立的Caco-2细胞模型可用于药物吸收机制研究。
Objective: To establish a human colon adenocarcinoma cell line (Caco-2) cell model and its drug research methods for preliminary study. Methods: The Caco-2 cell model was evaluated by measuring the transmembrane membrane resistance (TEER), the permeation of fluorescein and alkaline phosphatase (AKP). MTT assay was used to investigate the effect of HBSS buffer under different pH values, different temperatures and different time Toxicity to Caco-2 cells. Results: The measured values of each index met the requirements, indicating that the integrity, tightness and permeability of the Caco-2 cell model established in this experiment were good. For the study on the potential of Caco-2 cells for absorption for 4 h No effect on Caco-2 cell model. Conclusion: The established Caco-2 cell model can be used for drug absorption mechanism research.
