目的考察灵芝孢子油对慢性轻度不可预见刺激模型小鼠行为学影响及可能的神经生理机制。方法采用13种不同慢性轻度不可预见刺激对雄性BALB/C小鼠连续刺激建立小鼠抑郁模型。从第3周开始连续灌胃给予3个剂量的灵芝孢子油(850、283、141.5 mg·kg~(-1)),盐酸氟西汀(10 mg·kg~(-1))或空白溶媒(植物油)2周后,测定各组小鼠行为学变化,同时采用反相高效液相色谱法(RP-HPLC)测定海马谷氨酸(GLU)和γ-氨基丁酸(GABA),酶联免疫法测定脑源性神经营养因子(BDNF)的含量。结果与模型组比较,灵芝孢子油能增加抑郁小鼠的体重、糖水偏好率和旷场实验得分(P<0.01),缩短悬尾实验和强迫游泳实验的不动时间(P<0.05,P<0.01),明显降低海马GLU的含量(P<0.01),增加BDNF含量(P<0.01),但对GABA含量无影响(P>0.05)。结论灵芝孢子油对抑郁模型小鼠表现出明显的抗抑郁作用,其抗抑郁作用可能与其降低GLU和增加BDNF相关。
Objective To investigate the effects of Ganoderma lucidum spore oil on the behavior of chronic mild unpredictable stimulus model mice and its possible neurophysiological mechanism. Methods Thirteen different types of chronic mild unpredictable stimuli were used to establish a model of depression in male BALB / C mice. Three doses of ganoderma spore oil (850,283,141.5 mg · kg -1), fluoxetine hydrochloride (10 mg · kg -1) or blank vehicle (Vegetable oil) for 2 weeks, the behavioral changes of the mice in each group were measured and the contents of glutamate (GLU) and γ-aminobutyric acid (GABA) in the hippocampus were determined by RP-HPLC. Immunoassay was used to determine the content of brain-derived neurotrophic factor (BDNF). Results Compared with the model group, Ganoderma lucidum spores oil can increase body weight, sugar preference and open field test scores (P <0.01), shorten the hangover time and forced swimming time (P <0.05, P < 0.01), significantly decreased the content of GLU in hippocampus (P <0.01), increased BDNF content (P <0.01), but had no effect on GABA content (P> 0.05). Conclusion Ganoderma lucidum spore oil has obvious antidepressant effect on depression model mice, and its antidepressant effect may be related to its reduction of GLU and increase of BDNF.
