目的:应用超高压技术提取元胡中延胡索乙素,并结合正交试验优选最佳工艺。方法:在单因素实验的基础上,以延胡索乙素的提取率为考察指标,采用L8(27)正交试验,对影响超高压提取的主要因素(压力、时间、乙醇浓度、固液比)进行优选,并与回流提取、超声提取和微波提取进行比较。结果:超高压提取延胡索乙素的最佳工艺条件为:提取溶剂70%乙醇,压力250MPa,固液比1∶75(g∶mL),提取时间30 s;此提取条件下延胡索乙素的提取率为0.596 mg/g,分别高出回流提取、超声提取和微波提取25.2%、16.2%和18.0%。结论:超高压提取作为一种新兴的提取技术,具有提取温度低、提取率高、耗时短、能耗少等优点,为元胡中延胡索乙素的提取提供了一种全新的方法。
OBJECTIVE: To extract tetrahydropalmatine from Yuanhu by using ultrahigh-pressure technique and to optimize the optimum technology by orthogonal experiment. Methods: On the basis of single factor experiment, taking the extraction rate of tetrahydropalmatine as index, the main factors (pressure, time, ethanol concentration, solid-liquid ratio) Optimal, and compared with reflux extraction, ultrasonic extraction and microwave extraction. Results: The optimum extraction conditions for tetrahydropalmatine by ultra-high pressure were: extraction solvent 70% ethanol, pressure 250MPa, solid-liquid ratio 1:75 (g: mL), extraction time 30 s; extraction of tetrahydropalmatine The rates were 0.596 mg / g, 25.2%, 16.2% and 18.0% higher than reflux extraction, ultrasonic extraction and microwave extraction, respectively.
