目的:研究从山核桃叶中提取分离出来的黄酮单体2’,6’-二羟基-4’-甲氧基查耳酮对人脐静脉内皮细胞(HUVECs)与人单核细胞白血病细胞(THP-1)的黏附作用。方法:采用荧光标记检测经肿瘤坏死因子-α(TNF-α)刺激的HUVECs及THP-1之间黏附作用;采用荧光定量PCR及流式细胞技术,检测2’,6’-二羟基-4’-甲氧基查耳酮对HUVECs表面细胞黏附分子(VCAM-1)和细胞间黏附分子(ICAM-1)表达的影响。结果:2’,6’-二羟基-4’-甲氧基查耳酮能够明显减少TNF-α诱导的HUVECs与THP-1之间的黏附数量;能够抑制HUVECs表面黏附分子VCAM-1和ICAM-1的表达。结论:2’,6’-二羟基-4’-甲氧基查耳酮能够明显抑制TNF-α诱导的HUVECs与THP-1之间的黏附作用。降低HUVECs表面黏附因子VCAM-1、ICAM-1的表达可能是其抑制黏附作用的机制之一。
OBJECTIVE: To investigate the effect of 2 ’, 6’-dihydroxy-4’-methoxy chalcone, a flavone monomer extracted from the pecan tree leaves, on the proliferation of human umbilical vein endothelial cells (HUVECs) and human monocytic leukemia cells THP-1) adhesion. Methods: The adhesion between HUVECs and THP-1 stimulated by tumor necrosis factor-α (TNF-α) was detected by fluorescent labeling. Fluorescence quantitative PCR and flow cytometry were used to detect the expression of 2 ’, 6’-dihydroxy-4 Effect of methoxy chalcone on the expression of cell adhesion molecule (VCAM-1) and intercellular adhesion molecule (ICAM-1) on HUVECs. Results: 2 ’, 6’-dihydroxy-4’-methoxy chalcone significantly reduced the number of TNF-α-induced adhesion between HUVECs and THP-1, inhibited the expression of VCAM-1 and ICAM -1 expression. CONCLUSION: 2 ’, 6’-dihydroxy-4’-methoxy chalcone can significantly inhibit TNF-α-induced adhesion between HUVECs and THP-1. Decreasing the expression of adhesion molecules VCAM-1 and ICAM-1 on HUVECs may be one of the mechanisms of their inhibition of adhesion.
