目的观察不同检测试剂盒、不同抗原修复方法及修复液对乳腺癌环氧化酶-2(COX-2)表达的影响。方法①选取即用型和浓缩型COX-2单克隆抗体为70例乳腺癌组织标本进行免疫组化染色标记,浓缩型COX-2以1:80浓度稀释。②实验分为抗原未修复组和抗原修复组,抗原修复组又分别采用高温高压-EDTA缓冲液法(修复1组)、高温高压-枸橼酸缓冲液法(修复2组)、光波/微波-EDTA缓冲液法(修复3组)、光波/微波-枸橼酸缓冲液法(修复4组)做抗原修复处理。结果浓缩型COX-2的表达率明显比即用型COX-2高,且部分即用型阴性者浓缩型阳性,两者相比差异有统计学意义(χ2=25.3,P<0.01)。各抗原修复组染色效果比抗原未修复组好(P<0.01)。修复1组与修复3组、修复2组与修复4组相比,修复1组和修复2组抗原修复染色效果好,差异无统计学意义(P>0.05);修复1组与修复2组、修复3组与修复4组相比,修复1组和修复3组抗原修复染色效果好,差异有统计学意义(P<0.01)。结论即用型COX-2在乳腺癌中的表达存在假阴性,建议使用浓缩型COX-2抗体;光波/微波-EDTA缓冲液法修复COX-2抗原可明显提高免疫组化染色的敏感性,具有定位准确、阳性率高、操作简单、安全、组织结构保存好、不易掉片等优点。
Objective To observe the effect of different detection kits, different antigen retrieval methods and repair solution on the expression of cyclooxygenase-2 (COX-2) in breast cancer. Methods ① The samples of 70 cases of breast cancer were selected and labeled with COX-2 monoclonal antibody. The concentration of COX-2 was diluted by 1:80. The experiment was divided into antigen unmodified group and antigen retrieval group. The antigen retrieval group was treated by high temperature and high pressure-EDTA method (group 1), high temperature and high pressure-citrate buffer method (group 2), light / microwave -EDTA buffer method (repair three groups), light wave / microwave-citrate buffer method (repair four groups) do antigen repair treatment. Results The expression of COX-2 in concentrated form was significantly higher than that of ready-to-use COX-2, and some of the negative ones were positive in concentrated form. The difference was statistically significant (χ2 = 25.3, P <0.01). The staining effect of each antigen retrieval group was better than that of the non-antigen retrieval group (P <0.01). Repair 1 and repair 3 groups, repair 2 and repair 4 groups compared to repair 1 and repair 2 groups of antigen repair staining effect was good, the difference was not statistically significant (P> 0.05); repair 1 group and repair 2 groups, Compared with the repair group 4, the repair group 3 and repair group 3 antigen repair staining effect is good, the difference was statistically significant (P <0.01). Conclusions The expression of COX-2 in patients with breast cancer is false-negative, it is recommended to use concentrated COX-2 antibody. COX-2 antigen can be significantly improved by light / microwave- With accurate positioning, positive rate, simple, safe, well-preserved organizational structure, not easy to lose tablets and so on.
