利用甲基纤维素微量培养和~3H-TdR掺入法,研究了T淋巴细胞增殖动力学、辐射敏感性及T淋巴细胞集落形成单位(CFU-TL)。结果表明,外周血单个核细胞在一定浓度范围内,CFU-TL形成数随种植细胞数而增加,呈良好线性关系;培养12d的集落细胞在PHA-CM刺激下仍具有~3H-TdR掺入能力,但计数下降,表明培养后的集落细胞DNA合成能力减弱;γ射线辐照后,随剂量增加,集落数和~3H-TdR掺入数迅速降低,CFU-TL对辐射很敏感,D_0=2.3 Gy。
The proliferation kinetics, radiosensitivity, and T-lymphocyte colony forming units (CFU-TL) of T lymphocytes were studied using methyl cellulose microculture and ~3H-TdR incorporation method. The results showed that the number of CFU-TL increased with the number of cultured cells in a certain concentration range, showing a good linear relationship; 12-day cultured cells still had ~3H-TdR incorporation with PHA-CM stimulation. Ability, but the decrease in counts indicates that the DNA synthesis capacity of the colony cells after culture is weakened; after irradiation with γ-rays, the number of colonies and the number of ~3H-TdR incorporations decrease rapidly as the dose increases, CFU-TL is very sensitive to radiation, D_0= 2.3 Gy.
