以常绿萱草茎尖为外植体,研究了不同消毒方式、培养基配方对于外植体生长的影响,并探索了最适于愈伤组织形成、不定芽增殖和生根的培养基配方。结果表明:在无菌条件下,用75%酒精消毒30S,再转入0.1%升汞溶液浸泡8分钟消毒效果最好;有利于外植体产生愈伤组织的培养基配方是MS+2.0mg/LBA+0.2mg/LNAA;适合愈伤组织诱导生芽培养基配方是MS+1.0mg/L2.4-D+1.0mg/LKT;适合幼苗生根的培养基配方是MS+NAA0.5mg/L+活性炭0.2mg/L。
The stem tips of evergreen stems were used as explants to study the effects of different disinfection methods and medium formulations on the growth of explants. The optimal media for callus formation, adventitious bud multiplication and rooting were also explored. The results showed that under the condition of aseptic conditions, disinfection with 75% alcohol for 30s and then into 0.1% mercuric chloride solution for 8 minutes gave the best disinfection effect. The medium for the explants to produce callus was MS + 2.0mg /LBA+0.2mg/LNAA. The callus induction medium for callus induction was MS + 1.0mg / L 4-D + 1.0mg / LKT. The medium suitable for rooting was MS + NAA 0.5mg / L + Activated carbon 0.2mg / L.
