目的 :观察p2 1WAF1在高糖作用下腹膜间皮细胞肥大中的作用。方法 :用RT -PCR和WesternBlot法测定在高浓度葡萄糖 (含糖为 1 38%、3 86 % )作用 2 4h后体外培养的大鼠腹膜间皮细胞p2 1WAF1水平 ;用流式细胞仪分析细胞周期分布。结果 :高糖作用 2 4h后大鼠腹膜间皮细胞大多出现细胞肥大和停滞于细胞周期的G1期 ,以含糖 3 86 %组明显。p2 1WAF1mRNA和蛋白随着糖浓度增高表达增高 (含糖组间P <0 0 5 )。在和含糖组相同渗透压的甘露醇组和无血清常规培养基几乎无p2 1WAF1mRNA和蛋白的表达。结论 :p2 1WAF1可能在高糖作用下间皮细胞的肥大及G1期阻滞中起重要的调节作用
Objective: To observe the role of p2 1WAF1 in hypertrophy of peritoneal mesothelial cells under high glucose. Methods: The level of p21WAF1 in rat peritoneal mesothelial cells was determined by RT -PCR and Western blotting after 24 hours with high concentration of glucose (138%, 386%, respectively). The cells were analyzed by flow cytometry Periodic distribution. Results: Most of rat peritoneal mesothelial cells appeared hypertrophy and stagnated in the G1 phase of the cell cycle after 24 h of high glucose treatment, with a significant rate of 386%. The expression of p2 1WAF1 mRNA and protein increased with the increase of glucose concentration (P <0.05). There was almost no expression of p21WAF1 mRNA and protein in the mannitol group and the serum-free conventional medium at the same osmotic pressure as the sugar-containing group. CONCLUSION: p21WAF1 may play an important regulatory role in the hypertrophy of mesothelial cells and G1 arrest
