目的:研究甘草次酸钠(SG)对乳鼠心肌细胞的作用。方法:体外培养乳鼠心肌细胞,用放射免疫法和荧光法分别测定cAMP和[Ca~(2+)]_i。结果:SG 0.4mmol·L~(-1)于5,10和15min使心肌细胞搏动频率由73±9min~(-1)分别降至62±5,59±7和56±6min~(-1)。SG0.1和0.2mmol·L~(-1)分别在15和10,15min呈现以上相似的结果。SG0.2和0.4mmol·L~(-1)与心肌细胞37℃共孵10,15min,cAMP浓度和[Ca~(2+)]_i均被降低。SG 0.2mmol·L~(-1)处理组的cAMP含量低于对照组(1.09±0.18,1.12±0.35pmol per vial,P<0.05);[Ca~(2+)]_i的变化也类似(30±4nmol·L~(-1),P<0.01,28±6nmol·L~(-1)P<0.05)。SG 0.1和0.2mmol·L~(-1)提高心肌细胞悬液氧分压变化率(87%±5%,75%±4%,P<0.01),但SG0.4mmol·L~(-1)降低氧分压变化率(31%±2%,P<0.01)。结论:SG可保护心肌或治疗缺血性心脏病。
Objective: To study the effect of glycyrrhizin sodium (SG) on neonatal rat cardiomyocytes. Methods: Cardiomyocytes were cultured in vitro. The cAMP and [Ca ~ (2 +)] _i were measured by radioimmunoassay and fluorescence spectrometry. Results: The pulsatile frequency of cardiomyocytes was decreased from 73 ± 9min ~ (-1) to 62 ± 5,59 ± 7 and 56 ± 6min ~ (-1) after SG 0.4mmol·L -1 at 5, 10 and 15min respectively ). SG0.1 and 0.2mmol · L ~ (-1) showed similar results at 15 and 10,15min, respectively. SG0.2 and 0.4 mmol·L -1 were incubated with cardiomyocytes at 37 ° C for 10 and 15 min, respectively. Both cAMP concentration and [Ca 2+] i were decreased. The level of cAMP in SG 0.2 mmol·L -1 group was lower than that in control group (1.09 ± 0.18,1.12 ± 0.35 pmol per vial, P <0.05). The change of [Ca 2+] i was also similar 30 ± 4nmol·L -1, P <0.01, 28 ± 6nmol·L -1 P <0.05). SG 0.1 and 0.2 mmol·L -1 increased the rate of change of oxygen partial pressure in myocardial cell suspension (87% ± 5%, 75% ± 4%, P <0.01), but SG 0.4 mmol·L -1 ) Decreased the rate of change of oxygen partial pressure (31% ± 2%, P <0.01). Conclusion: SG can protect myocardium or treat ischemic heart disease.
