目的:分析宁波地区2004年-2009年流行的麻疹病毒分离株血溶素蛋白(Fusion protein,F)基因的序列特征。方法:采用RT-PCR扩增宁波地区2004年-2009年分离到的21株麻疹病毒株的F基因并进行序列测定,与疫苗株沪191、changchun-47、Edmonston株及国内核苷酸同源性最高的毒株Zhejiang05-04、IMB-1比较并分析毒株变异情况。结果:2004年-2009年宁波地区流行的21株麻疹病毒株中,核苷酸同源性为95.2%~100.0%,氨基酸同源性为98.0%~99.8%;与中国疫苗株沪191相比,其核苷酸同源性为95.2%~96.1%,氨基酸同源性为96.4%~97.3%。F基因3个糖基化位点(第32、64、70位AA),对病毒融合功能起重要作用的第112位、第121位、第195位、第549位氨基酸均未发生改变。结论:2004年-2009年宁波地区流行的麻疹病毒株F基因关键位点未发生变异,本研究对了解本地区麻疹病毒遗传变异规律及疫苗的选择具有重要意义。
Objective: To analyze the sequence characteristics of Fusion protein (F) gene of measles virus isolated in Ningbo from 2004 to 2009. Methods: F gene of 21 strains of measles isolates isolated in Ningbo area from 2004 to 2009 were amplified by RT-PCR and sequenced. The results showed that F gene was homologous to vaccine strains Shanghai 191, changchun-47, Edmonston and domestic nucleotide The highest strains Zhejiang05-04, IMB-1 comparison and analysis of strain variation. Results: The nucleotide sequences of 21 strains of measles isolates prevailing in Ningbo from 2004 to 2009 were 95.2% -100.0% and their amino acid identities were 98.0% -99.8%. Compared with the Chinese vaccine strain Shanghai 191 , The nucleotide homology was 95.2% ~ 96.1%, the amino acid homology was 96.4% ~ 97.3%. The three glycosylation sites (positions 32, 64 and 70 AA) of F gene had no change in the 112th, 121st, 195th and 549th amino acids that play an important role in virus fusion. Conclusion: There was no variation in the F gene between measles virus strains in Ningbo in 2004-2009. This study is of great significance in understanding the genetic variation of measles virus and the choice of vaccines in this area.
