外周血DC-CIK对人舌鳞癌细胞系Tca8113的抑瘤作用

目的:探讨体外致敏成熟树突状细胞(DC)与细胞因子诱导杀伤细胞(CIK)共培养细胞群对人舌鳞癌细胞系Tca8113移植瘤裸鼠的体内抑瘤效果。方法:人舌鳞癌外周血分离培养DC,体外诱导培养CIK细胞,将致敏成熟的DC与CIK体外扩增获得共培养细胞群DC-CIK细胞;于裸鼠单侧腋下接种Tca8113,将实验动物分为对照组A、实验组B和实验组C;接种后24h,A组移植瘤区注射生理盐水,B组于对侧腋下注射DC-CIK,C组同侧腋下注射DC-CIK,饲养4周;观察移植瘤的成瘤时间、成瘤率、生长曲线及组织学观察,采用SAS 6.12软件包对数据进行t检验、方差分析和χ2检验。结果:A组平均成瘤时间为7.63d,B组为9.5d,C组为12d,A、C组有显著差异(P=0.0132);注射后2周,A组成瘤率为100%,B组为87.5%,C组为62.5%,P>0.05;观察期内,从移植瘤生长曲线观察,A组瘤体增长最快,B组其次,C组最慢,A、B组无显著差异(P>0.05),A、C组差异显著(P=0.036)。结论:致敏DC-CIK共培养细胞对Tca8113移植瘤裸鼠有一定的体内抑瘤效应。 OBJECTIVE: To investigate the in vivo antitumor efficacy of cocultures of mature dendritic cells (DCs) and cytokine-induced killer cells (CIKs) in nude mice bearing human tongue squamous cell carcinoma cell line Tca8113 in vitro. Methods: DCs were isolated from peripheral blood of human tongue squamous cell carcinoma, DCs were cultured and induced in vitro. DCs and CIKs were obtained from DCs and CIKs which were matured in vitro. Tca8113 cells were inoculated into unilateral armpits of nude mice. The experimental animals were divided into control group A, experimental group B and experimental group C. After 24 hours of inoculation, the transplantation tumor area of ​​group A was injected with normal saline, while in group B, DC-CIK was injected into the contralateral armpit. CIK were fed for 4 weeks. The tumorigenic time, tumor formation rate, growth curve and histological observation of the transplanted tumor were observed. The data were analyzed by t test, variance analysis and χ2 test using SAS 6.12 software package. Results: The mean tumor formation time in group A was 7.63 days, in group B was 9.5 days, in group C was 12 days, there was a significant difference between groups A and C (P = 0.0132) Group A was 87.5%, Group C was 62.5%, P> 0.05; Observation of growth curves from the observation of tumor growth in Group A, the fastest growth of the tumor group B, followed by the C group was the slowest, A, B group no significant difference (P> 0.05). There was significant difference between A and C groups (P = 0.036). Conclusion: The sensitized DC-CIK co-cultured cells have certain in vivo antitumor effects on Tca8113 transplanted tumor in nude mice.
