目的探索脂多糖(lipolysaccharide,LPS)干预气道上皮细胞细胞因子的分泌及蛋白激酶C(protein kinase C,PKC)的表达。方法培养A549细胞株,0、0.1、1.0、10.0、20.0、30.0、50.0、100.0μg/mL的LPS干预A549细胞2、4、8、24、30、48 h以后收集细胞上清液及细胞质总蛋白,ELASA法检测A549细胞IL-1、IL-6、IL-8、IL-11的分泌,Western blot法检测相应的PKC的表达。结果不同浓度LPS干预A549细胞相同时间后细胞因子IL-1、IL-6、IL-8、IL-11的分泌量及PKC的表达量之间的差异均有统计学意义(P<0.01);PKC的表达和IL-11的分泌之间具有相关性(P<0.05)。结论LPS干预细胞因子IL-1、IL-6、IL-8、IL-11的分泌及PKC的表达,且LPS干预细胞因子IL-11的分泌可能和PKC信号通路有关。
Objective To investigate the effects of lipopolysaccharides (LPS) on the secretion of cytokines and the expression of protein kinase C (PKC) in airway epithelial cells. Methods A549 cells were cultured, A549 cells were treated with 0, 0.1, 1.0, 10.0, 20.0, 30.0, 50.0 and 100.0 μg / mL of LPS for 2, 4, 8, 24, 30, 48 h. Cell supernatants and cytoplasm The secretion of IL-1, IL-6, IL-8 and IL-11 in A549 cells was detected by ELASA method. The expression of PKC was detected by Western blot. Results The secretion of IL - 1, IL - 6, IL - 8 and IL - 11 and the expression of PKC in A549 cells treated with LPS at different concentrations for the same time had statistical significance (P <0.01). There was a correlation between PKC expression and IL-11 secretion (P <0.05). Conclusion LPS can upregulate the secretion of IL-1, IL-6, IL-8 and IL-11 and the expression of PKC, and the secretion of IL-11 may be related to PKC signaling pathway.
