目的:观察电针对帕金森病模型大鼠黑质区c-Jun氨基末端激酶(c-Jun N-terminal kinase,JNK)信号通路及炎症因子肿瘤坏死因子α(tumor necrosis factor-α,TNF-α)、γ-干扰素(interferon-γ,IFN-γ)、白细胞介素1β(interleukin-1β,IL-1β)的作用,探讨针刺治疗帕金森病的作用机制。方法:健康雄性SPF级SD大鼠32只,随机平均分为正常组、假手术组、模型组、电针组。模型组和电针组经颈背部注射鱼藤酮(1 mg/kg,溶于一定比例二甲亚砜和生理盐水中,浓度0.25 mg/m L)造模14 d。电针组给予电针“风府”(GV16)和“太冲”(LR3)两穴治疗,连续治疗14 d,其余各组不做治疗。假手术组给予相同剂量的二甲亚砜(Dimethyl sulfoxide,DMSO)和生理盐水混合液。采用免疫蛋白印迹法检测大鼠黑质区酪氨酸羟化酶(Tyrosine hydroxylase,TH)、磷酸化的c-Jun和TNF-α、IFN-γ、IL-1β蛋白表达情况。结果:与正常组、假手术组比较,模型组大鼠黑质区TH蛋白表达明显减少(P<0.05),p-c-Jun、TNF-α、IFN-γ、IL-1β蛋白表达明显增加(P<0.05);与模型组比较,电针组大鼠黑质区TH蛋白表达明显增加(P<0.05),p-c-Jun、TNF-α、IFN-γ、IL-1β蛋白表达明显减少(P<0.05)。结论:电针可以通过调节帕金森病模型大鼠体内MAPK/JNK信号通路,降低p-c-Jun在帕金森病模型大鼠黑质区的表达,进而减少炎症因子TNF-α、IFN-γ、IL-1β的表达,对帕金森病的发生发展起到一定的调节作用。
Objective: To observe the effects of electroacupuncture on c-Jun N-terminal kinase (JNK) signal pathway and the expression of tumor necrosis factor-α (TNF-α) in the substantia nigra of rats with Parkinson’s disease ), Interferon-γ (IFN-γ) and interleukin-1β (IL-1β), and explore the mechanism of acupuncture treatment of Parkinson’s disease. Methods: Thirty-two healthy male SPF SD rats were randomly divided into normal group, sham operation group, model group and electroacupuncture group. Rats in model group and EA group were injected rotenone (1 mg / kg dissolved in a certain proportion of dimethylsulfoxide and normal saline at a concentration of 0.25 mg / mL) for 14 days. The electroacupuncture group was given electroacupuncture “GV16” and “Taichong” (LR3) two points treatment for 14 consecutive days. The other groups were not treated. The sham operation group was given the same dose of dimethylsulfoxide (DMSO) and saline solution. The expression of tyrosine hydroxylase (TH), phosphorylated c-Jun, TNF-α, IFN-γ and IL-1β in rat substantia nigra was detected by Western blotting. Results: Compared with normal group and sham operation group, the expression of TH protein in substantia nigra of model group was significantly decreased (P <0.05) and the expression of pc-Jun, TNF-α, IFN-γ and IL- <0.05). Compared with the model group, the expression of TH protein in the substantia nigra of electroacupuncture group was significantly increased (P <0.05) and the protein expression of pc-Jun, TNF-α, IFN- 0.05). Conclusion: EA can decrease the expression of TNF-α, IFN-γ and IL-8 in the substantia nigra of Parkinson’s disease model by regulating MAPK / JNK signaling pathway in Parkinson’s disease model rats -1β expression, the occurrence and development of Parkinson’s disease play a regulatory role.
