目的探讨APOC-Ⅲ启动子C-482T和T-455C基因多态性与代谢综合征的关系。方法采用病例对照研究的方法,选择参与2010年浙江省代谢综合征患病率调查的180例病例及191例对照作为研究对象,应用直接测序法检测APOC-ⅢC-482T和T-455C的基因型,运用χ2检验比较其基因型及等位基因频率差异。结果病例组C-482TC/C、C/T和T/T基因型频率分别为28.89%、50.00%和21.11%,与对照组(36.60%,53.93%和10.47%)相比,差异有统计学意义(χ2=8.27,P<0.05),与C/C相比,C/T和T/T基因型OR值分别为1.14(95%CI:1.38~1.80)和2.48(95%CI:1.30~4.73);T-455C基因型频率与等位基因频率在病例组与对照组之间的差异无统计学意义(P>0.05)。结论 APOC-Ⅲ启动子C-482T基因多态性与MS及其组分高甘油三酯血症、超重和肥胖具有相关性,C/T和T/T基因型能够增加患MS的风险。
Objective To investigate the relationship between polymorphisms of APOC-Ⅲ promoter C-482T and T-455C gene and metabolic syndrome. Methods A case-control study was conducted in 180 cases and 191 controls participating in the survey of prevalence rate of metabolic syndrome in Zhejiang Province in 2010. The genotypes of APOC-Ⅲ C-482T and T-455C were detected by direct sequencing , Using χ2 test to compare the genotype and allele frequency differences. Results The frequencies of C-482TC / C, C / T and T / T genotypes were 28.89%, 50.00% and 21.11% respectively in the case group. The differences were statistically significant compared with the control group (36.60%, 53.93% and 10.47% (95% CI: 1.38 ~ 1.80) and 2.48 (95% CI: 1.30 ~ 1.80) in C / T and T / T genotypes compared with C / 4.73). There was no significant difference in the frequencies of genotype and allele of T-455C between the case group and the control group (P> 0.05). Conclusion C-482T polymorphism of APOC-Ⅲ promoter is associated with MS and its components hypertriglyceridemia, overweight and obesity. C / T and T / T genotypes may increase the risk of developing MS.
