目的观察转录因子Oct-4在ⅢB期结肠癌患者癌组织中的表达,并探讨Oct-4表达水平、部位同CD45RO~+淋巴细胞浸润密度之间的相关性,为结肠癌的诊断和判断预后提供依据。方法选取佳木斯大学附属第一医院65例经外科手术治疗切除的ⅢB期结肠癌患者的癌组织标本、36例癌旁组织标本及24例正常结肠组织标本。采用免疫组织化学染色法检测Oct-4,比较各组组织中Oct-4水平以及分布状况。同时,分析其与CD45RO+淋巴细胞浸润密度的关系。结果(1)Oct-4在结肠癌组织中高表达,在结肠癌旁组织中表达较弱,在正常结肠组织中无表达。(2)Oct-4蛋白表达中、强阳性的结肠癌组织中CD45RO+细胞浸润密度低于Oct-4水平阴性、弱阳性表达的组织。细胞核中表达Oct-4蛋白的结肠癌组织中CD45RO~+T淋巴细胞浸润密度高于核浆共表达的结肠癌组织。结论 Oct-4在ⅢB期结肠癌肿瘤细胞中表达,可作为诊断的参考指标;且Oct-4表达水平的高低可作为判断预后的标准。
Objective To observe the expression of Oct-4 in human cancerous tissue of stage ⅢB colon cancer, and to explore the correlation between the expression level of Oct-4 and the infiltration density of CD45RO ~ + lymphocytes, and to evaluate the prognosis of colon cancer Provide evidence. Methods Sixty-five surgically resected specimens of stage ⅢB colon cancer were collected from the First Affiliated Hospital of Jiamusi University, 36 specimens of paracancerous tissues and 24 specimens of normal colon tissues. Oct-4 was detected by immunohistochemical staining, and the levels and distribution of Oct-4 in each group were compared. At the same time, we analyzed its relationship with CD45RO + lymphocyte infiltration density. Results (1) Oct-4 was overexpressed in colon cancer tissues, weakly expressed in paracancerous tissues of colon and no expression in normal colon tissues. (2) In Oct-4 protein expression, the density of CD45RO + cells in strongly positive colon cancer tissues was lower than that in Oct-4 negative and weakly positive tissues. The infiltration density of CD45RO ~ + T lymphocytes in colon cancer tissues expressing Oct-4 protein in the nucleus was higher than that in colon nuclear carcinoma co-expressed. Conclusion The expression of Oct-4 in stage ⅢB colon cancer cells can be used as a reference marker for diagnosis. The expression level of Oct-4 may be used as a criterion for prognosis.
